Accumulation capacity of cadmium and copper and their effects on photosynthetic performance in Azolla filiculoides Lam. under induced rhizofiltration Gayana Botánica
Valderrama,Aly; Carvajal,Danny E.; Peñailillo,Patricio; Tapia,Jaime.
ABSTRACT This study evaluated the capacity of Azolla filiculoides to enhance heavy metal accumulation through the addition of different concentrations of cadmium (Cd) and copper (Cu), as ethylenediaminetetraacetic acid (EDTA) complexes, under hydroponic conditions. The physiological effect was determined by Photosynthetic efficiency (Fv/Fm) as an indicator of plant stress, and heavy metal content was measured with flame atomic absorption spectroscopy in the whole plants. The results obtained in rhizofiltration systems did not show an increase in accumulation when Cd-EDTA was added, but the Cu-EDTA increased the accumulation of Cu by plants. Although the Fv/Fm was decreased in all treatments, only the Cd-EDTA complex caused damage to photosystem II (PSII)...

Assessment of heavy metals in soils of a vineyard region with the use of principal component analysis Scientia Agricola
Valladares,Gustavo Souza; Camargo,Otávio Antônio de; Carvalho,José Ruy Porto de; Silva,Alessandra Maria Cia.
Agricultural management with chemicals may contaminate the soil with heavy metals. The objective of this study was to apply Principal Component Analysis and geoprocessing techniques to identify the origin of the metals Cu, Fe, Mn, Zn, Ni, Pb, Cr and Cd as potential contaminants of agricultural soils. The study was developed in an area of vineyard cultivation in the State of São Paulo, Brazil. Soil samples were collected and GPS located under different uses and coverings. The metal concentrations in the soils were determined using the DTPA method. The Cu and Zn content was considered high in most of the samples, and was larger in the areas cultivated with vineyards that had been under the application of fungicides for several decades. The concentrations of...

Assessment of micronutrient extractants from soils of Paraná, Brazil Rev. Bras. Ciênc. Solo
Sarto,Marcos Vinicius Mansano; Steiner,Fábio; Lana,Maria do Carmo.
In Brazil, plant-available micronutrients in the soil can be determined by several chemical extractants, the most common of which are dilute acid and chelating solutions. The purpose of this study was to assess the extractants 0.1 mol L-1 HCl, Mehlich-1, Mehlich-3 and DTPA for analysis of the micronutrients Cu, Zn, Fe, and Mn in soils from the state of Paraná. In samples from 12 soils (0-20 cm layer), wheat was planted (Triticum aestivum), grown for 42 days after emergence, and then bean (Phaseolus vulgaris) for 38 days. At the end of each planting period, the soil was sampled again. All extractants tested to assess the availability of Cu, Zn, Fe, and Mn correlated with each other. The efficiency of the extractants HCl, Mehlich-3 and DTPA in assessing...

Can pesticides, copper and seasonal water temperature explain the seagrass Zostera noltei decline in the Arcachon bay? ArchiMer
Gamain, Perrine; Feurtet-mazel, Agnes; Maury-brachet, Regine; Auby, Isabelle; Pierron, Fabien; Belles, Angel; Budzinski, Helene; Daffe, Guillemine; Gonzalez, Patrice.
Dwarf eelgrasses (Zostera noltei) populations have decreased since 2005 in Arcachon Bay (southwest France). Various stressors have been pointed out, however the role of xenobiotics like pesticides or copper (Cu) and of parameters like water temperature warming have not yet been explored. To determine their impact, Z. noltei individuals were collected in a pollution-free site and transferred to the laboratory in seawater microcosms. This dwarf eelgrass was exposed to a pesticide cocktail and copper, alone or simultaneously, at temperatures (10 °C, 20 °C, 28 °C) representative of different seasons. After a two-week contamination, leaf growth, leaf bioaccumulation of Cu, and differential expression of target genes were studied. Eelgrasses bioaccumulated Cu...

Changes in nitrate reductase activity and oxidative stress response in the moss Polytrichum commune subjected to chromium, copper and zinc phytotoxicity Braz. J. Plant Physiol.
Panda,Sanjib Kumar; Choudhury,Shuvasish.
The main aim of this paper was to investigate the effect of chromium (Cr), copper (Cu) and zinc (Zn) on nitrate reductase (NR) activity and oxidative stress responses in the moss Polytrichum commune. Cr, Cu and Zn resulted in the inhibition of NR activity. A decline in total chlorophyll content was observed after 24 and 48 h of metal treatment. Accumulation of the metals showed a dose and time dependent increase. High accumulation of Cu, Cr and Zn were seen in moss shoots after 24 and 48 h of treatment. Treatment of Cr, Cu and Zn for 24 or 48 h resulted in the increase of malondialdehyde (MDA) content in moss shoots. The highest increase was observed in shoots under Cu treatment followed by Cr and Zn. The MDA content was significantly higher after 48h....

Cloning and functional characterization of the gene encoding the transcription factor Acel in the basidiomycete Phanerochaete chrysosporium Biol. Res.
POLANCO,RUBÉN; CANESSA,PAULO; RIVAS,ALEXIS; LARRONDO,LUIS F; LOBOS,SERGIO; VICUÑA,RAFAEL.
In this report we describe the isolation and characterization of a gene encoding the transcription factor Acel (Activation protein of cup 1 Expression) in the white rot fungus Phanerochaete chrysosporium. Pc-acel encodes a predicted protein of 633 amino acids containing the copper-fist DNA binding domain typically found in fungal transcription factors such as Acel, Macl and Haal from Saccharomyces cerevisiae. The Pc-acel gene is localized in Scaffold 5, between coordinates 220841 and 222983. A S. cerevisiae acel null mutant strain unable to grow in high-copper medium was fully complemented by transformation with the cDNA of Pc-acel. Moreover, Northern blot hybridization studies indicated that Pc-acel cDNA restores copper inducibility of the yeast cup 1...

Combined effects of antifouling biocides on the growth of three marine microalgal species ArchiMer
Dupraz, Valentin; Stachowski-haberkorn, Sabine; Menard, Dominique; Limon, Gwendolina; Akcha, Farida; Budzinski, Helene; Cedergreen, Nina.
The toxicity of the antifouling compounds diuron, irgarol, zinc pyrithione (ZnPT), copper pyrithione (CuPT) and copper was tested on the three marine microalgae Tisochrysis lutea, Skeletonema marinoi and Tetraselmis suecica. Toxicity tests based on the inhibition of growth rate after 96-h exposure were run using microplates. Chemical analyses were performed to validate the exposure concentrations and the stability of the compounds under test conditions. Single chemicals exhibited varying toxicity depending on the species, irgarol being the most toxic chemical and Cu the least toxic. Selected binary mixtures were tested and the resulting interactions were analyzed using two distinct concentration-response surface models: one using the concentration addition...

COPPER CRISIS AND AGRICULTURAL RENAISSANCE IN ZAMBIA: AN ECONOMY-WIDE ANALYSIS AgEcon
Lofgren, Hans; Robinson, Sherman; Thurlow, James.
Zambia's strong dependence on copper exports has suppressed other tradables sectors, indicative of a Dutch disease phenomenon. The current copper crisis will have strong economic effects, possibly reversing such Dutch disease effects. We use a computable general equilibrium model built around a 1995 social accounting matrix to simulate the short- and long-run effects of two scenarios that reflect the current crisis, a 20 percent reduction in world copper prices and a complete collapse of copper mining. Compared to the short run, the long run is characterized by more flexibility in production technology and capital allocation. Both scenarios require a significant reduction in the "non-copper" trade deficit, absorption, and household consumption. The...

Copper desorption in a soil with variable charge Scientia Agricola
Casagrande,José Carlos; Jordão,Camila Beig; Alleoni,Luís Reynaldo Ferracciú; Camargo,Otávio Antônio de.
Adsorption processes of heavy metals in soils have been more extensively studied than desorption, in spite of this latter process being related to nutrient bioavailability in the soil solution. Copper desorption from surface (0-0.2 m) and subsurface (1.0 - 1.2 m) samples of an Anionic Acrudox was studied at two pH values (4.5 and 7.5). Soil samples were incubated with Cu rates varying from 0 to 400 mg kg-1, during 4 and 12 weeks, in the presence of CaCl2 as support electrolyte at concentrations of 0.01 and 0.001 mol L-1. Complete soil adsorption of added Cu was observed at pH 7.5 in all incubation periods, indicating that a 24h-shaking period was enough to reach equilibrium and maximum adsorption. Copper adsorption varied with the incubation period and was...

Copper improves the production of laccase by the white-rot fungus Pleurotus pulmonarius in solid state fermentation BABT
Tychanowicz,Giovana Kirst; Souza,Daniela F. de; Souza,Cristina G. M.; Kadowaki,Marina Kimiko; Peralta,Rosane Marina.
Pleurotus pulmonarius (Fr) Quélet, produced laccase as the main ligninolytic enzyme when cultivated on solid-state cultures using corn cob as substrate. The addition of copper greatly increased the production of enzyme. The addition of 25.0 mM CuSO4 increased the level of laccase from 270 to 1,420 U.L-1 and the fungus showed high resistance to copper under the conditions used in this work.

Copper Induced Lysosomal Membrane Destabilisation in Haemolymph Cells of Mediterranean Green Crab (Carcinus aestuarii, Nardo, 1847) from the Narta Lagoon (Albania) BABT
Aliko,Valbona; Hajdaraj,Gerta; Caci,Atenisa; Faggio,Caterina.
ABSTRACTDestabilisation of blood cell lysosomes in Mediterranean green crabCarcinus aestuarii was investigated using Neutral Red Retention Assay (NRRA). Crabs collected in Narta Lagoon, Vlora (Albania) during May 2014 were exposed in the laboratory to sub-lethal, environmentally realistic concentrations of copper. Neutral Red Retention Time (NRRT) and glucose concentration in haemolymph of animals were measured. The mean NRRT showed a significant reduction for the animals of the treatment group compared to the control one (from 118.6 ± 28.4 to 36.4 ± 10.48 min, p<0.05), indicating damage of lysosomal membrane. Haemolymph glucose concentration was significantly higher in the treatment group (from 37.8 ± 2.7 to 137.8.4 ± 16.2 mg/dL, p<0.05) than in...

Copper induces expression and methylation changes of early development genes in Crassostrea gigas embryos ArchiMer
Sussarellu, Rossana; Lebreton, Morgane; Rouxel, Julien; Akcha, Farida; Riviere, Guillaume.
Copper contamination is widespread along coastal areas and exerts adverse effects on marine organisms such as mollusks. In the Pacific oyster, copper induces severe developmental abnormalities during early life stages; however, the underlying molecular mechanisms are largely unknown. This study aims to better understand whether the embryotoxic effects of copper in Crassostrea gigas could be mediated by alterations in gene expression, and the putative role of DNA methylation, which is known to contribute to gene regulation in early embryo development. For that purpose, oyster embryos were exposed to 4 nominal copper concentrations (0.1, 1, 10 and 20 μg L−1 Cu2+) during early development assays. Embryotoxicity was monitored through the oyster embryo-larval...

Copper influence on polyphosphate metabolism of Cunninghamella elegans BJM
Souza,Patrícia Mendes de; Marinho,Petrusk Homero; Lima,Marcos Antônio Barbosa de; Nascimento,Aline Elesbão do; Takaki,Galba Maria de Campos.
The aim of this work was to evaluate the physiological aspects of polyphosphate metabolism of Cunninghamella elegans grown in presence of copper. The growth profile was obtained by means of biomass yields, orthophosphate consumption, polyphosphate accumulation and phosphatases activities. The results revealed the influence of copper on the growth, observed by biomass yields. Orthophosphate consumption was faster in cells grown in the presence of copper. The presence of copper in the culture medium induced polyphosphate accumulation. The polyphosphate level was almost constant in the beginning of control culture growth, and could be related to the exponential growth phase. On the other hand, the copper treated cultures exhibited a significant reduction in...

Copper levels evolution in Arcachon bay oysters : role of the regulation on antifouling paints ArchiMer
Alzieu, Claude; Barbier, Georges; Sanjuan, Jane.
The ban of tributyltin compounds in antifouling paints induced a strong decrease in the level of tin in Arcachon bay. At the same time, copper inputs have increased tenfold since 1982 owing to the widespread use of paints containing cuprous oxide. In spite of this large input, the statistical analysis of monitoring data (1979-1985) show a relative stability in the copper levels in Arcachon bay oysters.

Copper Stress on Photosynthesis of Black Mangle (Avicennia germinans) Anais da ABC (AABC)
GONZALEZ-MENDOZA,DANIEL; GIL,FRANCISCO ESPADAS y; ESCOBOZA-GARCIA,FERNANDO; SANTAMARIA,JORGE M.; ZAPATA-PEREZ,OMAR.
The effects of copper toxicity on the photosynthetic activities of Avicennia germinans was investigated using two CuSO4 concentrations (0.062 and 0.33 M) added in Hoagland's solution in an aerated hydroponic system. Photosynthesis and chlorophyll fluorescence were measured after 30 h of copper stress. Results obtained in this study show that increasing levels of Cu+2 of 0.062 and 0.33 M Cu+2 resulted in a general reduction of the stomatal conductance (28 and 18%, respectively) and 100% of inhibition of net photosynthesis. Additionally, at these concentrations of Cu+2, reductions of chlorophyll fluorescence parameters were also observed. These changes suggested that the photosynthetic apparatus of Avicennia germinans was the primary target of the Cu+2...
